چکیده

The occurrence of a negative energy balance (NEB) and concurrent metabolic changes may be responsible for impaired reproductive performance. A characteristic metabolic feature of NEB is the mobilization of body fat, reflected in an increased plasma non-esterified fatty acids (NEFA) concentration. High plasma concentration of NEFA is reflected in elevated NEFA follicular concentrations. The aim of the study was to examine the effects of different non-esterified fatty acids on oocyte and granulosa cell growth. Oocytes and granulosa cells were exposed to physiological and supra-physiological concentrations (in follicular fluid) of non-esterified fatty acids (Stearic acids:10, 20, 30 and 40μM or Palmitic acid: 20 40, 60 and 80μM or Oleic acid 40, 80, 120 and 160μM). The oocyte growth was measured in terms of the viability, maturation, cleavage, morulae/ blastocysts yield while granulosa cells growth was measured in terms of viability, cell number increment, cell metabolism, monolayer formation, support to oocyte growth and reversibility of functions after elimination of metabolic stressors (non-esterified fatty acids). We found that Oleic acid (40 and 80 μM) was beneficial to the oocyte and granulosa cell growth, Palmitic acids had no significant effect and stearic acid (30μM) was most inhibitory to oocyte and granulosa cell growth. The effect of different non-esterified fatty acids in combo levels on ovarian cell functions are in progress.

Effect of Metabolic Stress on Ovarian Activity and Reproductive Performance of Dairy Cattle: A Review

Reproductive efficiency in dairy cattle has been decreased over the last 5 decades despite significant gains in genetic selection, improved reproductive technologies and nutritional care. In high-yielding dairy cattle, the negative energy balance (NEB) during the first week post-partum adversely affects ovarian activity, mainly follicle growth and steroidogenesis. Circulating levels of non-este...

Lipotoxicity: impact on oocyte quality and reproductive efficiency in mammals

Lipotoxicity is characterized by excessive saturated fatty acids in the blood, increasing storage in non-adipose cells, which leads to changes in the expression pattern of genes related to endoplasmic reticulum stress (e.g., ATF4, ATF6, CHOP, and GRP78), proand anti-apoptotic pathways (e.g., Bax and Bcl-2, and protein stability, including heat shock proteins, e.g., HSP70). A negative sub-cellul...

P-29: Effects of Growth Factors and GranulosaCell Coculture on In vitro Maturation ofOocytes

Background: The maturation medium for in-vitro oocyte maturation is usually supplemented with serum. However, supplementation of serum from pregnant women adversely affects the outcome of in-vitro maturation. The purpose of the study was to assess if growth factors or granulosa cell coculture could overcome the adverse effects of pregnant women’s serum. Materials and Methods: The basal maturati...

Effect of Garcinia cambogia extract on body weight gain, feed intake and feed conversion ratio, and serum non-esterified fatty acids and C-reactive protein levels in rats fed with atherogenic diet

The aim of the study was to investigate the improving effect of Garcinia cambogia extract on performance metrics, and serum non-esterified fatty acids (NEFA) and C-reactive protein (CRP) levels in rats fed with atherogenic diet. Thirty, one-year-old, female Sprague-Dawley rats were randomly assigned to threeexperimental groups of ten animals each. Control group was fed with basal diet (2% liqui...

Fatty acid composition of the follicular fluid of normal weight, overweight and obese women undergoing assisted reproductive treatment: a descriptive cross-sectional study

BACKGROUND It has been well documented that the maturing oocyte is very vulnerable to changes in its micro-environment, the follicular fluid (FF). Recent research has focused on different components within this FF, like hormones, growth factors and metabolites, and how their concentrations are altered by diet and the metabolic health of the mother. It has been proposed that fatty acids (FAs) ar...
